The Rio Olympic Games-2016 silver medalist won her first game of the match in Tokyo 2020 in just 13 minutes with a score of 21-7.
She then went on to win the second set and the match 21-10.
World Number 58 Polikarpova seemed to struggle throughout the match with an injury.
Speaking about her first match, Sindhu said, "Even though my opponent was lower-ranked, I did not want to assume it would be easy. It is important to be focused. I made sure we had some rallies, and I got used to the court."
"It is important to make sure you play all your strokes and get used to them on the court. That's because you can't play them all of a sudden against a stronger opponent. It is important to know that your strokes are going well."
Sindhu next challenge will be overcoming the challenge from Hong Kong's world number 24 Cheung Ngan Yi.
Asked about the next contest, Sindhu said: "Each match is important. It is one match at a time. I am thinking of my next opponent. I will miss the spectators, but everyone is supporting me virtually," she said.
